What is epi-fluorescence and why is it better than trans-fluorescence?

Epi-fluorescence illuminates the specimen from above, through the objective lens itself — the same objective delivers excitation light and collects the fluorescence emission. That gives you a much brighter, higher-contrast image than trans-fluorescence (light from below), which is why it's the standard in modern fluorescence microscopy. All our fluorescence microscopes use this epi-fluorescence setup.

What fixation and staining do I need for fluorescence microscopy?

Common protocols that work with our SLM-FL-100: DAPI nuclear staining (fix cells with 4% paraformaldehyde, stain with DAPI at 0.1-1μg/ml), FITC-labelled antibody staining for immunofluorescence, acridine orange for live/dead discrimination, propidium iodide for dead cell staining.
